You have no choice to put the pedal to the metal on both runs and both cars needs to be different. What would you choose?
Uphill: Hennessey Venom GT! It's the fastest car in world. I would fly over the finish line right at the end.
Downhill: An MX5. Yes, I'd jump the car I know the most and see how fast it can go with the help of gravity!
So what would you opponauts choose?

Downhill - Lancia 037. Pure sex in car form. Group B rally heritage, AWD beating design, ferrari engine, RWD MR rally supercar.
Uphill - Fiesta ST. You can use ALL the power ALL the time and never have to worry about altitude holding you back. Incredibly well balanced, the chassis can use the torque of the little 1.6T very well. I had no issues nor lack of power when I drove up beartooth highway in the Rockies.

Hahaha, the downhill's tooooo easyyyyyyyy!
You bet I'll race a 20 ton monster! With a 12.1 Liter Diesel engine? YOU BET!
But the uphill was a challenge. I need TORQUE. Lots of it. And SPED.
So something like the Nismo Juke would be perfect! It's a crossover, so it'll do hills, and it has SPED. That, or I'm just being silly.

Uphill: Rallycross Fiesta. Acceleration that puts the Venom to shame and a suspension that ensures I can stay flat over those crests and land without destroying the car.
Downhill: Pro 2 off road truck. Similar logic. 900 horsepower and over two feet of suspension travel. Doesn't matter if I jump 200+ feet, won't be a problem.

Kept it realistic, so...
Headlights go up(hill): 991 GT3, because traction and dat noise.
Headlights go down(hill): S800, preferably with VFR1200 swap, cause light.

Uphill, C6 ZR1. The power, the noise, the air time.
Downhill, Solstice. Non-GXP, just the 177hp one. Probably similar reasons to you wanting to do the MX-5, but I don't own a Solstice. And I want to do it with the top down.
