My cities always succumb to gridlock, I have no idea how to keep traffic flowing in the middle of a 50k+ person city

Building interchanges in Skylines makes me feel like a masochist. I mean I hate doing it but once I started I need more.
I think it must been 6 hours just rebuilding them.
Also you have too many lanes. Never use more than you need.

Avoid intersections like the plague.
Have Highway access close to city centres as opposed to the edges.
Allow heavy traffic direct routes to their destinations.
Try to keep direct routes for everyone else to a minimum, it’s okay for Cims to double back on themselves.
Make liberal use of one way roads.
Use the smallest road possible.

I think I do everything wrong, then. My cities tend to be an antenna-like grid, with lots of dead-ends but frequent intersections on the major roadways. Maybe I’ll dust the game off sometime and take a whack at cleaning up the roads in one of my existing cities just for fun
